This competitive Enzyme-Linked Immunosorbent Assay (cELISA), supplied by Antibodies.com, is designed for the in vitro quantitative determination of rat Big Dynorphin in serum, plasma, tissue homogenates and other biological fluids. The assay employs the competitive inhibition enzyme immunoassay technique, in which a target-specific antibody has been pre-coated onto the microplate and Big Dynorphin present in samples or standards competes with biotin-labelled target for binding to that antibody. After washing, an HRP-avidin conjugate is added and TMB substrate is used to visualise the enzymatic reaction. The resulting colour intensity is inversely proportional to the Big Dynorphin concentration in the well.

The kit has a detection range of 7.81–500 pg/ml and a sensitivity of 3.3 pg/ml, with a total assay time of approximately 2 h 30 min. Detection is colorimetric, read at 450 nm on a microplate reader, using a pre-coated microplate arranged as 12 × 8-well strips. Validated recovery in neat samples (n=5) is reported at 82–102% for serum (average 92%), 91–100% for EDTA plasma (average 95%) and 88–100% for heparin plasma (average 94%). Kit components are stored at +4 °C or −20 °C as instructed, and freeze/thaw cycles should be avoided. The product is supplied for laboratory research use only.
