The assay uses a pre-coated microplate with a biotinylated anti-BTG1 detection antibody, HRP-avidin and TMB substrate, producing a yellow colour read colorimetrically at 450 nm on a microplate reader.

According to the manufacturer, the working range is 46.8–3000 pg/ml with a sensitivity of 16.38 pg/ml, and the total assay time is 4h 30m. Reported recovery averages fall between 84% and 91% across serum, EDTA plasma and heparin plasma (n=5 per matrix), with individual ranges spanning 80% to 102%. Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across the same matrices, with recoveries typically between 74% and 108%.
