Rat Beta-Site APP Cleaving Enzyme 1 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com for the in vitro quantitative determination of rat Beta-Site APP Cleaving Enzyme 1 (BACE1) in serum, plasma, cerebrospinal fluid or other biological fluids. The assay is run on a 96-well microplate (12 x 8 well strips) pre-coated with antibody specific for BACE1, employing a biotinylated anti-BACE1 detection antibody and HRP-Avidin conjugate.

Standards and samples are incubated in the wells and, after washing, TMB substrate produces a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm and concentration is calculated against a standard curve. Sensitivity is 0.062 ng/ml with an assay range of 0.156-10 ng/ml and a total assay time of approximately 4 hours 30 minutes.

Reported average recovery is 91% in serum (range 80%-102%, n=5), 90% in EDTA plasma (range 81%-100%, n=5), and 84% in heparin plasma (range 80%-89%, n=5). Linearity upon dilution (1:2 to 1:16) spanned 74%-108% across serum and plasma samples. The kit contains a lyophilized standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with components stored at +4°C or -20°C as instructed.
