Rat Bcl2 Associated X Protein ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay, supplied by Antibodies.com, is designed for the in vitro quantitative determination of rat Bax in serum, plasma, tissue homogenates or other biological fluids. An antibody specific for Bax has been pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, where Bax is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-Bax antibody is added, followed by an HRP-avidin conjugate. After further incubation and washing, TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of Bax captured in each well. The concentration of Bax is then calculated by reading the absorbance at 450 nm in a microplate reader and referring to a standard curve.

The reported assay range is 0.625-40 ng/ml with a sensitivity of <0.236 ng/ml. The platform consists of pre-coated 12 x 8 well strips, and the total assay time is 4h 30m. Sample recovery has been characterised in serum, EDTA plasma and heparin plasma, with average recoveries of 91% in serum (n=5, range 80%-102%), 90% in EDTA plasma (n=5, range 81%-100%) and 84% in heparin plasma (n=5, range 80%-89%). Dilution linearity has been examined at 1:2, 1:4, 1:8 and 1:16 dilutions across these three sample types. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
