Rat Aryl Hydrocarbon Receptor ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat Aryl Hydrocarbon Receptor. The kit is intended for use with tissue homogenates or other biological fluids and the result is read on a colorimetric microplate reader.

The assay uses a pre-coated microplate and a sandwich enzyme immunoassay technique. Standards and samples are added to the wells, where Aryl Hydrocarbon Receptor is captured by the immobilized antibody. After washing, a Biotinylated Anti-Aryl Hydrocarbon Receptor antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that changes to yellow after addition of stop solution; absorbance is read at 450 nm and the concentration is calculated from a standard curve. Sensitivity is <0.102 ng/ml and the assay range is 0.312-20 ng/ml. Total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.

Average recovery values (n=5) were 93% for serum (range 85-101%), 92% for EDTA plasma (range 86-99%) and 94% for heparin plasma (range 87-101%). Dilution linearity was demonstrated from 1:2 to 1:16 dilutions: serum (n=5) recoveries ranged 79-110%, EDTA plasma (n=5) ranged 73-109%, and heparin plasma (n=5) ranged 77-110%.
