Rat Arachidonate-5-Lipoxygenase ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is intended for the in vitro quantitative determination of rat 5 Lipoxygenase/5-LO. The kit is designed for use with tissue homogenates, cell lysates or other biological fluids, and shows reactivity with rat samples. The procedure uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and a colourimetric detection format read at 450 nm.

In the workflow, an antibody specific for 5 Lipoxygenase/5-LO is immobilised on the plate. Standards and test samples are added so that the analyte binds the immobilised antibody. After washing, a Biotinylated Anti-5 Lipoxygenase/5-LO Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue colour that changes to yellow on addition of an acidic stop solution. The intensity of the yellow product, read at 450 nm, is proportional to the amount of 5 Lipoxygenase/5-LO captured in each well.

The assay has a sensitivity of <0.35 ng/ml and an assay range of 0.781-50 ng/ml, with a total assay time of 4h 30m. Reported average recovery was 91% in serum, 90% in EDTA plasma and 84% in heparin plasma, and sample dilution linearity was demonstrated from 1:2 to 1:16. Kit components include a pre-coated microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with storage at +4°C or -20°C as instructed.
