A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Aquaporin 2 in tissue homogenates, cell lysates or other biological fluids. The assay employs a pre-coated microtiter plate, with standards and samples added to wells where Aquaporin 2 is captured by an immobilized antibody. Following incubation and washing, a Biotinylated Anti-Aquaporin 2 Antibody binds the captured target, and HRP-Avidin conjugate is then added. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Aquaporin 2 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The kit is reactive with rat samples and targets Aquaporin 2, also referred to by synonyms including AQP-2, AQP-CD, AQP2, Aquaporin-CD, Aquaporin-2 and collecting duct water channel protein. The assay has a sensitivity of <0.235 ng/ml and a working range of 0.625-40 ng/ml, with an assay time of approximately 4 hours 30 minutes. Detection is performed on a colorimetric microplate reader, and kit components are stored at +4°C or -20°C as instructed. Recovery performance has been demonstrated across serum, EDTA plasma and heparin plasma samples.
