Rat Apoptotic Peptidase Activating Factor 1 ELISA Kit

Manufactured for research laboratories, this Antibodies.com kit enables quantitative measurement of rat Apoptotic Peptidase Activating Factor 1 (APAF1) in tissue homogenates, cell lysates and other biological fluids. The assay uses a sandwich ELISA format on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with colorimetric detection read at 450 nm using a standard microplate reader. Total assay time is 4 hours and 30 minutes.

Reported performance characteristics include a sensitivity of <0.062 ng/ml and an assay range of 0.156–10 ng/ml. Standards and samples are captured by an APAF1-specific antibody pre-immobilised on the plate, then detected with a biotinylated anti-APAF1 antibody and an HRP-avidin conjugate. The reaction is visualised using TMB substrate, which produces a yellow product after addition of acidic stop solution; absorbance is proportional to APAF1 concentration and is interpolated from a standard curve.

Manufacturer-supplied sample data indicate average recovery of 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Reported linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
