Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat Apolipoprotein H (Apo-H) in serum, plasma or other biological fluids. An antibody specific for Apo-H is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, and following incubation and washing, a Biotinylated Anti-Apo-H Antibody is introduced, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; the intensity of the yellow colour is proportional to the amount of Apo-H captured. The concentration of Apo-H is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a reported sensitivity of 0.97 ng/ml and an assay range of 1.56-100 ng/ml, with a total assay time of 4h 30m. The platform is a pre-coated microplate supplied as 12 x 8 well strips, and detection is colorimetric using a microplate reader. Reported recovery values are: serum 80% - 102% (average 91%, n=5), EDTA plasma 81% - 100% (average 90%, n=5) and heparin plasma 80% - 89% (average 84%, n=5). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 was reported as 87-91%, 87-107%, 74-101% and 92-97% for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).

Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
