Rat Apolipoprotein B ELISA Kit

A Sandwich ELISA assay from Antibodies.com, designed for the in vitro quantitative determination of rat Apolipoprotein B in serum, plasma or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for Apolipoprotein B pre-coated onto a microtiter plate. Standards and test samples are added to the wells and Apolipoprotein B is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-Apolipoprotein B Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of Apolipoprotein B captured, and concentration is calculated by reading the O.D. absorbance at 450 nm and referring to the standard curve.

The assay has a sensitivity of <11.3 ng/ml and a measuring range of 31.2-2000 ng/ml, with a total assay time of 4h 30m. Detection is colourimetric and read on a microplate reader, using a pre-coated microplate platform of 12 x 8 well strips. Recovery performance has been validated in serum (n=5, average 91%, range 80-102%), EDTA plasma (n=5, average 90%, range 81-100%), and heparin plasma (n=5, average 84%, range 80-89%). Dilution linearity has also been demonstrated across the same sample types at 1:2, 1:4, 1:8, and 1:16 dilutions. Supplied components include a pre-coated 96 well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Kit components should be stored at +4°C or -20°C as instructed, and the product must not be used past its expiration date.
