Rat Amylase Alpha 1, Salivary ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com intended for the in vitro quantitative determination of rat Salivary alpha Amylase in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, in which an antibody specific for Salivary alpha Amylase has been pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells and, following incubation and washing, a Biotinylated Anti-Salivary alpha Amylase Antibody and an HRP-Avidin conjugate are applied. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of Salivary alpha Amylase captured in each well and is read as O.D. absorbance at 450 nm.

The kit is read on a colorimetric microplate reader with a sensitivity of <0.28 ng/ml and an assay range of 0.781-50 ng/ml. Total assay time is 4h 30m. Sample recovery has been validated for serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, 81%-100%, average 90%) and heparin plasma (n=5, 80%-89%, average 84%). Dilution linearity has been examined at 1:2, 1:4, 1:8 and 1:16 dilutions across these sample types.
