A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rat Aminolevulinate Delta Synthase 1 (Alas1) in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for Alas1 has been immobilised. Standards and test samples are added to the wells, followed by washing and incubation with a Biotinylated Anti-Alas1 Antibody that binds captured Alas1. An HRP-Avidin conjugate is then added, and a TMB substrate solution is used to visualise the enzymatic reaction, producing a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm with a colorimetric microplate reader and Alas1 concentration is calculated from the standard curve.

The reported assay range is 0.781–50 ng/ml with a sensitivity of <0.36 ng/ml. Total assay time is 4 hours 30 minutes. Kit components are stored at +4 °C or −20 °C as instructed and the kit should not be used past its expiration date.

Average recovery values for rat samples (n=5) are reported as 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity is provided for serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions, with reported ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma respectively.
