Rat Aminolevulinate Delta Dehydratase ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat ALAD (aminolevulinate delta dehydratase) in serum, plasma, tissue homogenates, cell lysates or other biological fluids. An antibody specific for ALAD is pre-coated onto a 96-well microplate. Standards and samples are added to the wells, where ALAD is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-ALAD Antibody binds the captured ALAD. An HRP-Avidin conjugate is then added; after further incubation and washing, TMB substrate solution is applied and catalysis yields a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm and ALAD concentration calculated against the standard curve.

The assay has a sensitivity of <0.128 ng/ml, a quantitative range of 0.312–20 ng/ml, and a total assay time of 4h 30m. Reported average recovery values are: serum (n=5) 80%–102% (average 91%), EDTA plasma (n=5) 81%–100% (average 90%) and heparin plasma (n=5) 80%–89% (average 84%). Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed; the kit must not be used past its expiration date.
