From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of rat alpha 1 Antitrypsin in serum, plasma or other biological fluids.

An antibody specific for alpha 1 Antitrypsin is pre-coated onto the wells of a 96-well microplate. Standards and samples are added, and after incubation the wells are washed and incubated with Biotinylated Anti-alpha 1 Antitrypsin Antibody followed by HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow on addition of stop solution, and the absorbance is read at 450 nm. The quoted assay range is 78.1-5000 ng/ml with a sensitivity of 29 ng/ml, and the total assay time is 4h 30m.

Validation data (n=5) show average recovery of 91% from serum (range 80%-102%), 90% from EDTA plasma (81%-100%) and 84% from Heparin plasma (80%-89%). Dilution linearity (n=5) at 1:2, 1:4, 1:8 and 1:16 dilutions was 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for Heparin plasma.
