A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rat alanine aminotransferase (also referred to as alanine transaminase) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips); an antibody specific for the target is immobilised on the plate and captures analyte from standards and test samples. Following washing, a biotinylated anti-alanine aminotransferase antibody binds captured analyte, and an HRP-avidin conjugate completes the detection step. TMB substrate is then added; catalysis generates a blue colour that converts to yellow on addition of acidic stop solution, and the absorbance is read at 450 nm in a colorimetric microplate reader.

The standard curve covers a measuring range of 0.781–50 ng/ml with a sensitivity of <0.28 ng/ml. Total assay time is 4h 30m. Recovery validation performed on n=5 samples gave ranges of 80%–102% (serum, average 91%), 81%–100% (EDTA plasma, average 90%) and 80%–89% (heparin plasma, average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions produced recoveries of 87%–91%, 87%–107%, 74%–101% and 92%–97% for serum (n=5); 90%–105%, 84%–101%, 90%–101% and 79%–108% for EDTA plasma (n=5); and 84%–95%, 92%–105%, 82%–105% and 89%–91% for heparin plasma (n=5). Supplied components include the pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers; kit components should be stored at +4°C or -20°C as indicated and the kit should not be used past its expiration date.
