Designed for the in vitro quantitative determination of rat PACAP-38 (Pituitary Adenylate Cyclase Activating Polypeptide 1) in serum, plasma, tissue homogenates, or other biological fluids, the Antibodies.com Rat PACAP-38 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA). The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips, using a biotinylated anti-PACAP-38 detection antibody followed by an HRP-avidin conjugate and a TMB substrate; absorbance is read at 450 nm on a colorimetric microplate reader. The quantitative measurement range is 6.25-400 pg/ml with a sensitivity of <3.12 pg/ml, and the total assay time is approximately 4 hours 30 minutes.

Reported spike-and-recovery performance in serum ranged from 80%-102% (average 91%), in EDTA plasma from 81%-101% (average 91%), and in heparin plasma from 80%-89% (average 84%), each across five samples. Dilution linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across the same sample types. Supplied components include the pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, to be stored at +4°C or -20°C as instructed and not used past the expiration date.
