Rat Adenylate Cyclase 10, Soluble ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat ADCY10/SAC in tissue homogenates or other biological fluids. The assay employs an antibody specific for ADCY10/SAC pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, where the target is captured by the immobilised antibody. After washing, a Biotinylated Anti-ADCY10/SAC Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. The concentration of ADCY10/SAC is calculated by reading the absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a sensitivity of <0.125 ng/ml and a quantitative range of 0.312-20 ng/ml, with colourmetric detection on a pre-coated microplate comprising 12 x 8 well strips. Total assay time is 4h 30m. Recovery has been characterised in serum (average 91%, n=5), EDTA plasma (average 94%, n=5) and heparin plasma (average 95%, n=5) across serial dilution factors of 1:2, 1:4, 1:8 and 1:16. Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
