Rat Activity Regulated Cytoskeleton Associated Protein ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Activity Regulated Cytoskeleton Associated Protein (Arc) in tissue homogenates or other biological fluids. Reactivity: Rat.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for Arc is pre-coated onto a 96-well microplate. Standards and samples are added, and Arc present in each sample binds to the immobilised antibody. After washing, a Biotinylated Anti-Arc Antibody is added, which binds the captured Arc, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Arc captured in each well, and the concentration of Arc can be calculated by reading the O.D. absorbance at 450 nm against the standard curve.

The kit has a sensitivity of <0.051 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Detection is colourimetric on a pre-coated microplate (12 x 8 well strips). Kit components should be stored at +4°C or -20°C as instructed. Mean recovery is 91% in serum (range 80%–102%, n=5), 90% in EDTA plasma (range 81%–100%, n=5) and 84% in heparin plasma (range 80%–89%, n=5).
