Rat Actin Alpha 1, Skeletal Muscle ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of rat alpha Skeletal Muscle Actin in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Distributed by Antibodies.com, the kit employs the sandwich immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for alpha Skeletal Muscle Actin is immobilised on the plate; standards and samples are added and captured analyte is detected using a Biotinylated Anti-alpha Skeletal Muscle Actin Antibody and an HRP-Avidin conjugate. TMB substrate produces a yellow product whose absorbance at 450 nm is proportional to analyte concentration, measured on a colorimetric microplate reader.

Reported performance includes a measuring range of 0.156–10 ng/ml with a sensitivity of <0.067 ng/ml. Mean recovery (n=5) was 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Linearity at dilutions 1:2, 1:4, 1:8 and 1:16 was demonstrated in serum (70–107%, 89–93%, 86–104%, 80–103%), EDTA plasma (95–100%, 85–110%, 81–99%, 90–102%) and heparin plasma (77–104%, 88–95%, 94–104%, 80–107%), each n=5. Total assay time is 4h 30m. Components include a pre-coated plate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers, stored at +4°C or −20°C as instructed.
