Rat Acid Phosphatase 5, Tartrate Resistant ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com is designed for the in vitro quantitative determination of rat Tartrate Resistant Acid Phosphatase in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips); standards and samples are added, the target is captured by the immobilised antibody, and a biotinylated anti-Tartrate Resistant Acid Phosphatase detection antibody is applied, followed by an HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of the acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader and Tartrate Resistant Acid Phosphatase concentration calculated from the standard curve.

The kit reports a sensitivity of <0.29 ng/ml and a working range of 0.781–50 ng/ml, with a total assay time of 4h 30m. Spiking recovery was 80%–102% (average 91%, n=5) in serum, 81%–100% (average 90%, n=5) in EDTA plasma, and 80%–89% (average 84%, n=5) in heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Supplied components include a pre-coated 96-well microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers; components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
