The assay uses an antibody specific for ADAMTS4 pre-coated onto a microtiter plate; standards and samples are added and the captured ADAMTS4 is detected using a Biotinylated Anti-ADAMTS4 Antibody followed by an HRP-Avidin conjugate. A TMB substrate produces a blue product that turns yellow after addition of acidic stop solution, and the absorbance is read at 450 nm in a microplate reader with the concentration calculated against a standard curve.

According to the manufacturer, the assay has a product range of 3.12–200 ng/ml, a sensitivity of 1.15 ng/ml, and a total assay time of 4 hours 30 minutes. Recovery data are reported for serum (range 80%–102%, average 91%, n=5), EDTA plasma (range 81%–101%, average 91%, n=5), and heparin plasma (range 80%–89%, average 84%, n=5).
