A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat ADAM9 in tissue homogenates, cell lysates, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for ADAM9 pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, with ADAM9 bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-ADAM9 Antibody binds the captured ADAM9, and an HRP-Avidin conjugate is added. TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of ADAM9 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The kit has a sensitivity of 0.054 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of approximately 4 hours 30 minutes. The platform is a pre-coated microplate supplied as 12 x 8 well strips, with colourimetric detection. Kit components include lyophilised standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and plate sealers.
