The assay uses a pre-coated 96-well microplate read on a colorimetric microplate reader, with absorbance measured at 450 nm. The assay range is 0.312–20 ng/ml and the reported sensitivity is <0.112 ng/ml, with a total assay time of 4h 30m. In the sandwich ELISA workflow, ADAM17 in standards or samples is captured by an immobilised specific antibody, detected with a biotinylated anti-ADAM17 antibody, and visualised using an HRP-avidin conjugate and TMB substrate. The resulting yellow product has a density proportional to ADAM17 concentration, which is calculated from the standard curve.
