The assay uses an anti-ADAM10 antibody pre-coated onto the microplate wells. Standards and samples are bound, washed, and incubated with a Biotinylated Anti-ADAM10 Antibody, followed by an HRP-Avidin conjugate. TMB substrate produces a yellow-coloured product whose optical density is proportional to ADAM10 concentration and is interpolated from a standard curve.

The assay range is 78.1-5000 pg/ml with a sensitivity of <27 pg/ml. Sample recovery (n=5) averages are 91% for serum (range 83-100%), 94% for EDTA plasma (range 90-98%) and 95% for heparin plasma (range 89-102%). Dilutional linearity at 1:2, 1:4, 1:8 and 1:16 (n=5): serum 83-110%, 79-100%, 90-98%, 82-108%; EDTA plasma 90-98%, 90-105%, 73-109%, 87-91%; heparin plasma 88-103%, 77-105%, 95-98%, 87-110%.
