Rat 11-Beta-Hydroxysteroid Dehydrogenase Type 1 ELISA Kit

A sandwich enzyme-linked immunosorbent assay from Antibodies.com, designed for the in vitro quantitative determination of rat 11-Beta-Hydroxysteroid Dehydrogenase Type 1 (HSD11B1) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips). An antibody specific for HSD11B1 is immobilised on the plate; standards and samples are added and the target is captured, followed by incubation with a Biotinylated Anti-HSD11B1 Antibody and HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to HSD11B1 concentration, read as absorbance at 450 nm on a colourimetric microplate reader.

Reported sensitivity is <0.123 ng/ml with a measuring range of 0.312-20 ng/ml, and the total assay time is 4 hours 30 minutes. Average recovery across sample types was 91% in serum (n=5; range 83-99%), 90% in EDTA plasma (n=5; range 80-100%), and 90% in heparin plasma (n=5; range 82-99%). Linearity across 1:2 to 1:16 dilutions was 74-109% in serum, 78-110% in EDTA plasma, and 77-109% in heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
