Rabbit Ubiquitin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of rabbit Ubiquitin. The kit is reactive with rabbit samples and is intended for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses the sandwich enzyme immunoassay technique. An antibody specific for Ubiquitin is pre-coated onto the wells of a microplate. Standards and test samples are added, and Ubiquitin present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-Ubiquitin Antibody is added and binds the captured analyte. HRP-Avidin conjugate is then added, followed by TMB substrate solution. Catalysis produces a blue-coloured product which changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and Ubiquitin concentration is determined from the standard curve.

Performance characteristics reported by the manufacturer include a product range of 3.12–200 ng/ml and a sensitivity of 0.78 ng/ml, with a total assay time of 4 h 30 min. Recovery was evaluated (n = 5) in serum at 80–102% (average 91%), EDTA plasma at 81–99% (average 90%), and heparin plasma at 80–89% (average 84%). Sample dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions in serum, EDTA plasma and heparin plasma (n = 5 each). Kit components are stored at +4 °C or -20 °C as instructed; the kit should not be used past the expiration date.
