Rabbit Selectin, Platelet ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of rabbit P Selectin in serum, plasma, tissue homogenates or other biological fluids. An antibody specific for P Selectin is pre-coated onto the microtiter plate. Standards and test samples are added to the wells, where P Selectin present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-P Selectin Antibody is added, followed by an HRP-Avidin conjugate and further washing. TMB substrate solution is then added; catalysis by HRP produces a blue product that turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the quantity of P Selectin captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay is configured as a quantitative sandwich ELISA with colorimetric readout on a pre-coated microplate, with a stated assay range of 0.625–40 ng/ml and sensitivity of <0.282 ng/ml. The total assay time is 4 hours 30 minutes. Reagent storage instructions specify +4°C or -20°C as indicated on the components, and the kit should not be used past its expiration date. The intended instrument is a colorimetric microplate reader.

Reported validation performance includes recovery averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma samples.
