Description

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of inducible nitric oxide synthase (iNOS) in rabbit samples. The kit uses an antibody specific for iNOS that is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, where iNOS binds to the immobilized capture antibody; a biotinylated anti-iNOS detection antibody is then introduced, followed by an HRP-Avidin conjugate. TMB substrate produces a colorimetric signal read at 450 nm, and iNOS concentration is determined by reference to the standard curve.

The assay has a stated sensitivity of 0.54 ng/ml and a product range of 1.56-100 ng/ml, with a total assay time of 4h 30m. It is intended for use with tissue homogenates, cell lysates and other biological fluids. Reported recovery ranges were serum (n=5) 80%-102% with an average of 91%, EDTA plasma (n=5) 81%-100% averaging 90% and heparin plasma (n=5) 80%-89% averaging 84%. Dilution linearity was reported at 1:2 to 1:16 for serum (87-91%, 87-107%, 74-101%, 92-97%), EDTA plasma (90-105%, 84-101%, 90-101%, 79-108%) and heparin plasma (84-95%, 92-105%, 82-105%, 89-91%). Kit components are stored at +4°C or -20°C as instructed.

An additional sandwich ELISA kit for rabbit iNOS, validated with serum, plasma and tissue homogenate samples, has been documented with a sensitivity of <0.188 ng/mL, a range of 0.313-20 ng/mL and a 4-hour protocol, and is reported to show no cross-reactivity with other analogs or variants of iNOS.
