Rabbit N-Terminal Pro Brain Natriuretic Peptide ELISA Kit

Antibodies.com provides a sandwich ELISA kit designed for the in vitro quantitative determination of rabbit N-Terminal Pro Brain Natriuretic Peptide (NT-ProBNP) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate in a 12 x 8 well strip format and is completed in 4h 30m. The kit is reactive with rabbit samples.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for NT-ProBNP is pre-coated onto the plate; standards and samples are added and the analyte is captured by the immobilized antibody. After washing, a Biotinylated Anti-NT-ProBNP Antibody binds the captured NT-ProBNP, followed by addition of an HRP-Avidin conjugate. TMB substrate is then added; catalysis produces a blue-coloured product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of NT-ProBNP captured. Absorbance is read at 450 nm using a colorimetric microplate reader and the concentration is calculated by referring to the standard curve.

Sensitivity is less than 11.3 pg/ml and the assay range is 31.2-2000 pg/ml. Recovery was assessed: in serum (n=5) the average recovery was 91% across a range of 80%-102%; in EDTA plasma (n=5) average recovery was 91% across 81%-102%; in heparin plasma (n=5) average recovery was 84% across 80%-89%. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 gave ranges of 87-91%, 87-107%, 74-101% and 92-97% in serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% in EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% in heparin plasma (n=5). Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
