From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (sELISA) is intended for the in vitro quantitative determination of rabbit Matrix Metalloproteinase 2 (MMP2) in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate onto which an antibody specific for MMP2 has been immobilised. Standards and test samples are added to the wells, where MMP2 binds to the captured antibody. After washing, a biotinylated anti-MMP2 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate solution is then added; catalysis by HRP produces a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of MMP2 captured, and concentration is calculated by reading the OD absorbance at 450 nm on a colorimetric microplate reader, referencing the standard curve.

The kit reports a sensitivity of <1.35 pg/ml and an assay range of 3.12-200 pg/ml, with a total assay time of 4 hours 30 minutes. Validation recovery data shows serum (n=5) ranging 80%-102% with an average of 91%, EDTA plasma (n=5) ranging 81%-100% with an average of 90%, and heparin plasma (n=5) ranging 80%-89% with an average of 84%. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions was: serum (n=5) 87-91%, 87-107%, 74-101%, 92-97%; EDTA plasma (n=5) 90-105%, 84-101%, 90-101%, 79-108%; heparin plasma (n=5) 84-95%, 92-105%, 82-105%, 89-91%.
