Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rabbit Matrix Metalloproteinase 1 (MMP1) in serum, plasma, or other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for MMP1. Standards and test samples are added to the wells, where MMP1 present in each sample is bound by the immobilised capture antibody. Following incubation and washing, a Biotinylated Anti-MMP1 Antibody is added, which binds the captured MMP1, and an HRP-Avidin conjugate is subsequently introduced. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of MMP1 captured in each well, and the concentration of MMP1 is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Quantitative readout is performed on a colorimetric microplate reader. The product range is 31.2–2000 pg/ml with a sensitivity of <13.8 pg/ml, and the total assay time is 4h 30m. Kit components include a Pre-Coated 96 Well Microplate (12 x 8 well strips), Lyophilised Standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and Plate Sealers (5 adhesive strips). Components are stored at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.

Recovery has been validated across matrices: serum (range 80–102%, average 91%, n=5), EDTA plasma (range 81–100%, average 90%, n=5), and heparin plasma (range 80–89%, average 84%, n=5). Linearity has also been assessed at dilutions of 1:2, 1:4, 1:8, and 1:16 across the same sample types.
