Rabbit Interleukin 8 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com for the in vitro quantitative determination of rabbit interleukin-8 (IL-8) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips, with a capture antibody specific for IL-8 immobilised on the wells. Standards and samples are added; captured IL-8 is detected with a Biotinylated Anti-IL-8 Antibody and an HRP-Avidin conjugate, and the signal is developed with TMB substrate to produce a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and is directly proportional to IL-8 concentration.

The quantitative assay has a working range of 15.6–1000 pg/ml and a sensitivity of <6.7 pg/ml, with a total assay time of 4h 30m. Spike-and-recovery validation returned an average of 94% (range 87%–101%) in serum (n=5), 95% (range 90%–100%) in EDTA plasma (n=5) and 96% (range 91%–102%) in heparin plasma (n=5). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 was confirmed in serum (82–101%), EDTA plasma (80–102%) and heparin plasma (79–104%).
