Rabbit Interleukin 10 ELISA Kit

An in vitro quantitative sandwich ELISA designed for the measurement of rabbit IL-10 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The Antibodies.com assay uses a 96-well microplate pre-coated with an antibody specific for IL-10. Standards and samples are added to the wells, and following incubation and washing, a biotinylated anti-IL-10 antibody and an HRP-Avidin conjugate are applied in sequence. TMB substrate is then added, producing a blue-coloured product that turns yellow after addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the IL-10 concentration is calculated against the standard curve.

The standard curve covers a product range of 78.1–5000 pg/ml with a sensitivity of <24 pg/ml, and the full procedure takes 4 h 30 min. Spike-and-recovery values across serum, EDTA plasma and heparin plasma samples (n=5) averaged 91%, 96% and 95% respectively, with corresponding ranges of 83–100%, 90–103% and 89–102%. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was generally within 73–110% across the three sample matrices.

Kit components include a pre-coated 96-well microplate (12 x 8-well strips), lyophilized standard, detection solution A, detection solution B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
