A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rabbit IL-1 beta in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit employs a 96-well microtiter plate pre-coated with an antibody specific for IL-1 beta; standards and samples are added to the wells and IL-1 beta present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-IL-1 beta Antibody is added, followed by HRP-Avidin conjugate and TMB substrate solution. The HRP enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution, and the density of yellow is proportional to the amount of IL-1 beta captured in each well. The concentration of IL-1 beta is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.
The assay is quantitative and colourimetric, run on a pre-coated microplate of 12 x 8 well strips, with a total assay time of 4h 30m. Reported performance includes a product range of 78.1-5000 pg/ml and a sensitivity of <33 pg/ml. Sample recovery has been characterised in serum (n=5, range 83%-100%, average 91%), EDTA plasma (n=5, range 90%-103%, average 96%) and heparin plasma (n=5, range 89%-102%, average 95%), with linearity assessed across dilutions of 1:2, 1:4, 1:8 and 1:16. Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
