Description

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of rabbit Hepatocyte Growth Factor (HGF) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for HGF is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where HGF is captured by the immobilised antibody. After washing, a Biotinylated Anti-HGF Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise HRP enzymatic activity, producing a blue product that turns yellow upon addition of acidic stop solution. HGF concentration is determined from the O.D. absorbance measured at 450 nm in a colourimetric microplate reader, by reference to the standard curve. The product range is 15.6–1000 pg/ml and the sensitivity is <6.1 pg/ml, with a total assay time of 4h 30m.

Components supplied include Lyophilised Standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 Adhesive Plate Sealers. Storage conditions range from +4°C to –20°C depending on the component. Reported spike-and-recovery ranges are: Serum 80–102% (average 91%, n=5), EDTA Plasma 81–100% (average 90%, n=5) and Heparin Plasma 80–89% (average 84%, n=5). Linear dilution recovery across 1:2, 1:4, 1:8 and 1:16 dilutions was 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5).
