Rabbit Endothelin 1 ELISA Kit

This competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of rabbit Endothelin 1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the competitive enzyme immunoassay technique on a pre-coated 96-well microplate (12 x 8 well strips). During incubation, Endothelin 1 present in samples or standards competes with the fixed amount of immobilised antigen for binding sites on a biotinylated anti-Endothelin 1 antibody; bound complex is detected with an HRP-Avidin conjugate and visualised using TMB substrate, producing a blue product that changes to yellow after addition of acidic stop solution. The density of yellow is inversely proportional to the amount of Endothelin 1 present, and the concentration is calculated from the O.D. absorbance read at 450 nm in a colorimetric microplate reader.

The assay has a stated product range of 15.6-1000 pg/ml and a sensitivity of <5.63 pg/ml, with a total assay time of 2h 30m. Kit components (lyophilised standard, detection solution A, detection solution B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers) are stored at +4°C or -20°C as instructed. Reported average recovery (n=5) is 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%). The kit should not be used past its expiration date.
