Rabbit Apolipoprotein E ELISA Kit

A competitive Enzyme-Linked Immunosorbent Assay (cELISA) kit from Antibodies.com, intended for the in vitro quantitative determination of rabbit Apolipoprotein E in serum, plasma or other biological fluids. The assay employs a competitive enzyme immunoassay technique on a 96-well microtiter plate pre-coated with Apolipoprotein E antigen. During incubation, Apolipoprotein E present in samples or standards competes with the immobilised antigen for binding sites on a Biotinylated Anti-Apolipoprotein E Antibody; the greater the analyte concentration in a well, the less antibody that binds to the plate.

After washing to remove unbound antibody, an HRP-Avidin conjugate is added to the wells. A TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of an acidic stop solution. The intensity of the yellow colour is inversely proportional to the Apolipoprotein E concentration, which is calculated by reading the optical density at 450 nm in a colorimetric microplate reader and referring to the standard curve. The kit has a reported sensitivity of <0.30 ng/ml and a working range of 0.625–40 ng/ml, with a total assay time of 2 h 30 min.

Reported recovery values are 80–102% (average 91%) for serum (n=5), 81–100% (average 90%) for EDTA plasma (n=5) and 80–89% (average 84%) for heparin plasma (n=5). Linearity is also reported across dilutions of 1:2, 1:4, 1:8 and 1:16 for each of these sample types. Kit components are intended for storage at +4°C or -20°C as instructed in the manual and should not be used past the expiration date.
