Progesterone ELISA Kit

The Universal Progesterone ELISA Kit from Antibodies.com is a competitive Enzyme-Linked Immunosorbent Assay (cELISA) designed for the in vitro quantitative determination of Progesterone in serum, plasma, tissue homogenates, and other biological fluids. The assay carries a universal reactivity profile and is reported to be specific for Progesterone, with no cross-reactivity noted against other analogs or variants.

The procedure uses a pre-coated microplate, with detection carried out colorimetrically by measuring absorbance at 450 nm using a microplate reader. The complete assay takes 2 hours. During the run, Progesterone in standards or samples competes with immobilized Progesterone on the plate for binding to an assay-specific antibody. After washing, an HRP-Streptavidin conjugate is added, followed by TMB substrate. The enzymatic reaction produces a blue product that turns yellow upon addition of the acidic stop solution, with colour intensity inversely proportional to the Progesterone concentration. The kit is shipped at +4°C, and components should be stored at +4°C or -20°C as instructed in the protocol manual; freeze/thaw cycles should be avoided and the kit must not be used past its expiration date.

The standard curve covers a product range of 0.313–20 ng/mL, with a sensitivity of 0.188 ng/mL. In validation work, serum samples (n=10) showed recovery of 89–104%, averaging 95%. EDTA plasma (n=10) gave 90–101% with a 95% average, while heparin plasma (n=10) yielded 89–101% with a 96% average. Dilution linearity was also assessed at 1:2, 1:4, and 1:8 dilutions across all three sample matrices.
