Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of porcine Vascular Cell Adhesion Molecule 1 (VCAM1) in serum, plasma, tissue homogenates and other biological fluids. The format is a 96-well microplate pre-coated with an antibody specific for VCAM1; standards and samples are added to the wells and captured VCAM1 is detected using a Biotinylated Anti-VCAM1 Antibody followed by HRP-Avidin conjugate and TMB substrate solution, producing a blue product that turns yellow after the addition of acidic stop solution. The intensity of the yellow colour is proportional to VCAM1 concentration and is read at 450 nm in a colorimetric microplate reader.

The assay has a reported range of 15.6–1000 ng/ml and a sensitivity of <6.7 ng/ml, with a total assay time of 4h 30m. Spike-and-recovery has been validated in serum (n=5, range 80%–102%, average 91%), EDTA plasma (n=5, 81%–100%, average 90%) and heparin plasma (n=5, 80%–89%, average 84%). Linearity has been examined at 1:2, 1:4, 1:8 and 1:16 dilutions for each sample type. Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
