Porcine Tumor Necrosis Factor Beta ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of porcine LTA (Tumor Necrosis Factor Beta) in serum, plasma, or other biological fluids. The microplate is pre-coated with an antibody specific for LTA; standards and samples are added so that the analyte binds to the immobilised antibody. Following incubation and washing, a biotinylated anti-LTA antibody is added, followed by HRP-avidin conjugate and TMB substrate, and the colour change is read at 450 nm against a standard curve.

Detection is colorimetric on a pre-coated 96-well microplate arranged as 12 × 8 well strips, with a sensitivity of <5.1 pg/ml and an assay range of 15.6–1000 pg/ml. The protocol runs in approximately 4 hours 30 minutes. In validation testing with five replicates per matrix, mean recoveries were 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%), and 84% in heparin plasma (range 80–89%).

Supplied components include a pre-coated 96-well microplate, two vials of lyophilised standard, detection solutions A and B, 30× wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, with individual storage instructions ranging from +4°C to −20°C.
