Porcine Tumor Necrosis Factor Alpha ELISA Kit

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of porcine TNF alpha in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) and follows a sandwich immunoassay format with a biotinylated anti-TNF alpha detection antibody, HRP-avidin conjugate and TMB substrate. The colour change is read at 450 nm on a colorimetric microplate reader, with the intensity of the yellow product proportional to TNF alpha concentration, calculated against the standard curve.

The standard product range spans 62.5–4000 pg/ml with a sensitivity of <27.8 pg/ml, and the total assay time is 4 hours 30 minutes. Reported recovery in spiked samples averaged 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Linearity across 1:2 to 1:16 dilutions was 87–91% for serum, 90–105% for EDTA plasma and 84–95% for heparin plasma (all n=5). Kit contents include lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at +4°C or –20°C as instructed and not used past the kit expiration date.
