Porcine Trypsin ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of porcine Trypsin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique using a pre-coated 96-well microplate (12 x 8 well strips) bearing an antibody specific for Trypsin. Standards and test samples are added to the wells; Trypsin present in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-Trypsin Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate solution is then added; HRP-catalysed conversion produces a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Trypsin captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

Stated performance characteristics include a product range of 1.56–100 ng/ml and a sensitivity of 0.58 ng/ml (summarised elsewhere as <0.58 ng/ml), with a total assay time of approximately 4 h 30 min. Spike-and-recovery testing gave: serum (n=5) 80–102%, average 91%; EDTA plasma (n=5) 81–99%, average 90%; heparin plasma (n=5) 80–89%, average 84%. Dilution linearity at sample dilutions of 1:2, 1:4, 1:8 and 1:16 was reported as: serum (n=5) 87–91%, 87–107%, 74–101%, 92–97%; EDTA plasma (n=5) 90–105%, 84–101%, 90–101%, 79–108%; heparin plasma (n=5) 84–95%, 92–105%, 82–105%, 89–91%. Kit components are stored at +4 °C or -20 °C as instructed, and the product must not be used past its expiration date.
