Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of porcine Transforming Growth Factor Beta Induced Protein (TGFBI) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The kit uses a 96-well microtiter plate pre-coated with a TGFBI-specific antibody. Standards and test samples are added, and TGFBI in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-TGFBI Antibody is added, followed by HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of TGFBI captured, and concentration is calculated by reading the O.D. absorbance at 450 nm against the standard curve.

Read on a colorimetric microplate reader, the assay has a stated sensitivity of <0.23 ng/ml, a measurement range of 0.937–60 ng/ml and a total run time of 4h 30m. Reported recoveries (n=5) were 80%–102% (average 91%) for serum, 81%–100% (average 90%) for EDTA plasma, and 80%–89% (average 84%) for heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was reported as 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
