This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of porcine Transferrin in serum, plasma or other biological fluids. The kit is reactive with porcine samples and is read on a colorimetric microplate reader. It employs the sandwich enzyme immunoassay technique, with an antibody specific for Transferrin pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, and the immobilized antibody captures Transferrin present in each sample.

Following incubation and washing, a Biotinylated Anti-Transferrin Antibody is added, which binds to the captured Transferrin in each well. An HRP-Avidin conjugate is then added and the plate incubated. TMB substrate solution is used to visualise the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Transferrin captured in each well, and the concentration of Transferrin can be calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The reported assay range is 0.312-20 ng/ml, with a sensitivity of <0.105 ng/ml. The total assay time is 4 hours 30 minutes, and the detection type is colourimetric on a pre-coated microplate of 12 x 8 well strips. Reported recovery averages are 91% for serum, 90% for EDTA plasma and 84% for heparin plasma (n=5). Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
