From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of porcine TIMP2 in serum, plasma, tissue homogenates or other biological fluids. The assay employs a 96-well microplate pre-coated with a TIMP2-specific antibody, with bound target detected using a biotinylated anti-TIMP2 antibody, an HRP-avidin conjugate and TMB substrate. The enzymatic reaction is stopped with an acidic solution producing a yellow colour, and absorbance is read at 450 nm on a colorimetric microplate reader. The standard curve covers 0.312-20 ng/ml, with a reported sensitivity of <0.103 ng/ml, and the full incubation protocol runs in approximately 4 hours 30 minutes.

Spike-and-recovery performance has been characterised across sample matrices, with average recoveries of 91% in serum (range 80%-102%, n=5), 90% in EDTA plasma (range 81%-100%, n=5) and 84% in heparin plasma (range 80%-89%, n=5). Dilution linearity has been evaluated at 1:2, 1:4, 1:8 and 1:16 across the same matrices, with serum recoveries of 87-91%, 87-107%, 74-101% and 92-97% respectively; EDTA plasma recoveries of 90-105%, 84-101%, 90-101% and 79-108%; and heparin plasma recoveries of 84-95%, 92-105%, 82-105% and 89-91%.
