Porcine Tissue Inhibitors Of Metalloproteinase 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of porcine TIMP1 in serum, plasma, tissue homogenates and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for TIMP1, configured as 12 x 8 well strips. Captured TIMP1 is detected with a biotinylated anti-TIMP1 antibody followed by an HRP conjugate, and the enzymatic reaction is visualised with TMB substrate, producing a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of TIMP1 is calculated from a standard curve. Total assay time is approximately 4 to 4.5 hours, and the kit is supplied with a lyophilised standard, detection reagents, wash and sample dilution buffers, TMB substrate, stop solution and plate sealers.

Sample validation data are provided for serum, EDTA plasma and heparin plasma, with recovery reported across multiple dilutions. According to the manufacturer's specifications, one kit format reports a sensitivity of less than 13.6 pg/ml and an assay range of 31.2–2000 pg/ml, while another reports a sensitivity of less than 0.188 ng/ml and an assay range of 0.313–20 ng/mL. Cross-reactivity is reported as specific for TIMP1, with no reported cross-reactivity with other analogs or variants.
