Porcine Tissue Factor ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of porcine Tissue Factor in plasma. The assay uses a pre-coated 96-well microplate to which an antibody specific for Tissue Factor has been immobilised. Standards and test samples are added to the wells, and the captured Tissue Factor is detected using a Biotinylated Anti-Tissue Factor Antibody followed by HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of Tissue Factor captured. Concentration is calculated by reading O.D. absorbance at 450 nm against a standard curve.

The reported assay range is 0.156–10 ng/ml and sensitivity is <0.061 ng/ml, with a total assay time of 4 hours 30 minutes and colourimetric detection on a microplate reader. Recovery data are provided for serum (average 92%, range 82–102%, n=5), EDTA plasma (average 95%, range 90–100%, n=5) and heparin plasma (average 94%, range 88–100%, n=5). Linearity is reported at dilutions of 1:2, 1:4, 1:8 and 1:16: serum (n=5) ranges 86–110%, 77–102%, 90–96% and 84–108%; EDTA plasma (n=5) 88–100%, 90–103%, 76–110% and 86–93%; heparin plasma (n=5) 89–102%, 75–106%, 94–97% and 88–110%.

Kit components include a pre-coated microplate (12 x 8 well strips), lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or –20°C as instructed. The kit is intended for plasma samples and should not be used past the expiration date.
