This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is intended for the in vitro quantitative determination of porcine Surfactant Protein A (PSAP) in serum, plasma, tissue homogenates, lung lavage fluid or other biological fluids.

The assay uses a pre-coated microplate onto which an antibody specific for Surfactant Protein A/PSAP has been immobilised. Standards and test samples are added to the wells, and any Surfactant Protein A/PSAP present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Surfactant Protein A/PSAP Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of the acidic stop solution. The absorbance is read at 450 nm in a microplate reader and the concentration calculated against the standard curve. The stated measuring range is 3.12–200 ng/ml with a sensitivity of <1.16 ng/ml. Total assay time is approximately 4h 30m and detection is colourimetric.

Reported average recovery values are 80–102% in serum (n=5), 81–99% in EDTA plasma (n=5) and 80–89% in heparin plasma (n=5). The kit is supplied with a pre-coated microplate, lyophilised standard, Detection Solutions A and B, a 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
