Description

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of porcine Stromal Cell Derived Factor 1 (SDF1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for SDF1. Standards and test samples are added to the wells and any SDF1 present is captured by the immobilized antibody. After washing, a Biotinylated Anti-SDF1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of SDF1 captured, and the concentration is calculated from the O.D. absorbance read at 450 nm in a microplate reader, referenced to a standard curve.

The kit has a standard range of 0.625-40 ng/ml and a sensitivity of <0.245 ng/ml, with the full assay taking approximately 4h 30m. Detection is colorimetric on a pre-coated microplate of 12 x 8 well strips. Kit components should be stored at +4°C or -20°C as indicated and the kit should not be used past its expiration date.

Spike-and-recovery data (n=5) show ranges of 80-102% for serum (average 91%), 81-100% for EDTA plasma (average 90%), and 80-89% for heparin plasma (average 84%). Dilution linearity (n=5) at 1:2, 1:4, 1:8 and 1:16 gives 87-91%, 87-107%, 74-101% and 92-97% respectively for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
