Porcine Plasminogen Activator, Urokinase ELISA Kit

Antibodies.com supplies a Porcine Plasminogen Activator, Urokinase ELISA Kit (uPA) configured as a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of porcine uPA in serum, plasma or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for uPA; standards and test samples are added, followed by a Biotinylated Anti-uPA Antibody and an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate solution, producing a blue-coloured product that changes to yellow on addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader. The density of yellow is proportional to the amount of uPA captured in each well.

The reported assay range is 0.156–10 ng/ml and the sensitivity is <0.054 ng/ml. Total assay time is 4h 30m. Kit components should be stored at +4°C or −20°C as instructed and the product should not be used past its expiration date. The assay is suitable for serum and plasma samples.

Recovery performance has been characterised across sample types. In neat samples, average recovery ranges were 82%–102% for serum (n=5), 91%–103% for EDTA plasma (n=5) and 86%–101% for heparin plasma (n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 gave recovery ranges of 89%–107%, 72%–106%, 89%–92% and 88%–108% for serum; 84%–102%, 90%–99%, 81%–110% and 82%–98% for EDTA plasma; and 90%–100%, 80%–108%, 91%–97% and 90%–107% for heparin plasma.
